c2 P22 under TetR repressible promoter

Consists of c2 P22 coding region and TetR repressible promoter. When TetR is present, the production of c2 P22 will be stopped. When both TetR and aTc are present, there will be production of c2 P22. Also when there is no TetR, c2 P22 will be transcribed.

Comment

To make this part, we ligated R0040 to P0353. Strangely enough, according to the sequencing results, our part contains R0040 but only part of C0053 (coding sequence C2 P22) that is present within P0353.
