The Improved Fusarium Toehold Switch Sequence was derived from NUPACK and was constructed with the second lowest normalized ensemble defect rate possible. The sequence forms a hairpin loop, hiding machinery necessary for translation. Improved Fusarium Toehold Switch Sequence The Improved Fusarium Toehold Switch Sequence was derived from NUPACK and was constructed with the second lowest normalized ensemble defect rate possible. The sequence forms a hairpin loop, hiding machinery necessary for translation. However, in the presence of the FRP1 gene of Fusarium oxysporum, a plant pathogen, the hairpin loop unravels, allowing for expression of the downstream gene to occur. As a result, this sequence can be paired with a reporter gene to function as a biosensor for the presence of the plant disease F. oxysporum.
