The bistable switch is composed of trc promoter(BBa_K3805208), P2 promoter(BBa_I746104), lacI- LVA (BBa_C0012)and mcherry-ompt(BBa_K3805886).When AIP is released, it will act on P2 promoter and start the expression of lacI-LVA and downstream mcherry-ompt. LacI-LVA inhibits trc promoter.When the concentration of AIP decreases, the P2 promoter will be closed, and the inhibition of lacI-LVA on the trc promoter will be relieved.The trc promoter starts the expression of the target protein.

LacI-LVA inhibits trc promoter.As the concentration of AIP decreases, the downstream transcription of P2 promoter will be down-regulated and the inhibition of lacI-LVA on the trc promoter will thus be lifted.The trc promoter then initiates the its downstream expression.

Usage and Biology
