araBAD promoter from Salmonella Typhimurium
This is the native promoter of the Salmonella Typhimurium ara-operon. The ara-operon encodes genes for the breakdown of arabinose. Its expression is regulated by the AraC, which binds the PBAD in absence of arabinose. In the presence of arabinose, the AraC dimer inhibiting expression is released and the expression of downstream genes is allowed.1
As a part of our project "TargetTaxi", we aim to use this promoter to regulate the expression of the SPI-1 master regulator, hilA. Thereby, allowing the overexpression of SPI-1 type 3 secretion machinery and its assembly at the cell poles. The presence of the machinery at the cell poles will result in its presence in the membrane of minicells.
