This gene cassette has been designed to be a part of the genetic circuit for an inducible kill switch in Bacillus subtilis. The gene coding for the XylR repressor ( protein is downstream to the P43 constitutive promoter of Bacillus subtilis (BBa_K143013) and RBS for Bacillus subtilis for constitutive expression. The xylR gene is upstream to porcine teschovirus-1 P2A Peptide Linker for effective transcription. Downstream to the linker, a reporter gene sfGFP (BBa_K3889002) has been placed to check the working of the cassette in Bacillus subtilis.
