{˂GFP-LVA˃} Fast-Degrading GFP

The purpose of this part is to encode a fast degrading GFP. Wildtype GFP typically have a half-life that may last for several bacterial generations. However, the addition of an AANDENYALAA tag to the C-terminal of the wildtype GFP will decrease this halflife to 40 minutes (Andersen et al, 1998). By changing the last three residues of this tag, various fast-degrading GFP were created. GFP[LVA] has a halflife of 40 minutes (Miller et al., 2000).

Usage of a fast degrading GFP may act as a better reporter of changes in gene transcription over a period of time.
