flhB promoter followed by its natural RBS
flhB is a class 2 gene that is part of the construction of the flagella ( Ordering Genes in a Flagella Pathway by Analysis of Expression Kinetics from Living Bacteria - S. Kalir, J. McClure, K. Pabbaraju, C. Southward, M. Ronen, S. Leibler, M. G. Surette, U. Alon ).
Its promoter is consitutively repressed.
It is activated by FlhD4C2 and maybe by FliA with different strength ( Using a Quantitative Blueprint to Reprogram the Dynamics of the Flagella Gene Network - Shiraz Kalir and Uri Alon ).
WARNING
The promoter cloned here corresponds to the promoter region given in the following paper:
Kalir, S. et al. Ordering genes in a flagella pathway by analysis of expression kinetics from living bacteria. Science 292, 2080-2083 (2001)
Position 1964392-1964190 in MG1655 genome sequence.
Note that the promoter region given here includes the beginning of the flhB protein. It thus cannot be used with any protein coding biobrick since it will add a small peptide and most probably produce a frame shift in your protein. To use it in front of an ORF, this ORF must be preceded by a stop codon in the right frame.
We are currently building a new version of this promoter which will include stop codons so that it will comply with the biobrick standard and be usable in front of any protein coding biobrick.
