This is an intermediate part of the oscillatory system which can be used to build our final modular biosensor BBa_K3440021. LuxR which is originally from Vibrio Fischeri (UniProtKB - P12746), is constitutively expressed under the J23100 promoter. In the presence of 3OC6-HSL (a lactone produced by LuxI from Vibrio Fischeri, UniProtKB - P12747), the LuxR:3OC6-HSL complex can activate Plux, which controls production of MtrB. This oscillatory part will be present in Shewanella oneidensis MR-1 mtrb knockout.
Due to the pandemics, we haven’t been able to use biobricks to create the iGEM Stockholm 2020 parts. Those parts were ordered as gene blocks from Integrated DNA Technologies Inc.. As a result, the sequences of the biobricks used are the same, but the scars between biobricks might differ, as well as the final size of the part.

As this part is an intermediate in making the final oscillatory system it cannot be characterized individually unless added into BBa_K3440021.

MtrB under Lux promoter

Pconst(BBa_J23100) - RBS(BBa_B0034) - LuxR(BBa_C0062) - DT(BBa_B0015) - Plux(BBa_R0062) - RBS(BBa_B0034) - mtrb(BBa_K3146001) - DT(BBa_B0015)

Usage and Biology

As this part is an intermediate in making the final oscillatory system it cannot be characterized individually unless added into BBa_K3440021.
