Reticulate Binding Protein 5

P. falciparum Reticulocyte Binding Protein Homologue 5 (PfRH5) is part of a parasite protein family, the reticulocyte-binding protein homologue (RH) which mediates host-parasite interaction. PfRH5 is the only member of the family necessary for the erythrocyte invasion in all tested strains, by binding with the erythrocyte surface protein basigin[1][2][3]. PfRH5 is a member of the super family of erythrocyte ligands referred to as the Reticulocyte Binding Like proteins (RBLs), at least one member of which is found in every Plasmodium genome[1] PfRH5 is unique even in the PfRH family in that it is much smaller compared to the other proteins of the family and that the gene encoding for PfRH5 is resistant to genetic deletion in all tested strains, showing its importance in blood-stage growth[2].

PfRH5 folds such that two three-helical bundles come together in a kite-like formation, and the resultant structure presents binding sites for basigin and inhibitory antibodies at one tip[4]. Basigin binds at the tip of PfRH5, distant from the flexible loop and C-terminus, with both domains and the intervening linker directly contacting PfRH5. Most of the contact area occurs through hydrogen bonds between the backbone of strands A and G of the basigin N-terminal domain and loops at the tip of PfRH5.[4]
