Sequencing of the transformation of part BBa_K2974500 confirmed the toehold construct’s function when it was aligned with the desired toehold sequence. In addition, the results for the toehold was confirmed through a dual plasmid transformation in BL21 E.coli cells. The dual plasmid transformation presented greater fluorescence measured in lux values in comparison to the dual plasmid transformation with part BBa_K2974700, a toehold construct with a strong promoter (BBa_J23100). The biobricked toehold's (BBa_K2974500) function has been validated through experimentation and sequencing.

Medium Promoter (BBa_J23106) Toehold GFP
Sequencing of the transformation of part BBa_K2974500 confirmed the toehold construct’s function when it was aligned with the desired toehold sequence. In addition, the results for the toehold was confirmed through a dual plasmid transformation in BL21 E.coli cells.
