This part includes the -35 and -10 elements of the strong pTac promoter. The human c-Myc promoter was found to have a sequence capable of forming an i-motif in its nuclease hypersensitive element III. This i-motif forming sequence was modified to fit on the template strand, around the -35 region of the pTac promoter.
