Psp1w1 promoter has a single operator located between the -35 and -10 elements, where TAL Effector 1 can tightly bind, decoupling its expression from plasmid/gene copy number. Copy number positively affects GOI expression, while its repressor, being under the same positive influence, compensates for this change resulting in the disassociation of the GOI’s expression from the plasmid’s copy number
