Insulated Psp1w1 promoter (B0032 rbs)

Psp1w1 B0032 sfGFP

Short Description This composite part was submitted by iGEM Thessaloniki and consists of Psp1w1 promoter driving the expression of sfGFP fluorescent marker. Psp1w1 promoter has a single operator located between the -35 and -10 elements, where TAL Effector 1 can tightly bind, decoupling its expression from plasmid/gene copy number. Copy number positively affects GOI expression, while its repressor, being under the same positive influence, compensates for this change resulting in the disassociation of the GOI’s expression from the plasmid’s copy number

Psp1w1 (B0032) device leads to a stronger sfGFP expression when compared to Psp1w1 B0030 device (BBa_2839008) due to the different RBS. RiboJ insulates the translational unit from surrounding sequences. This part is included in our Part Collection, a genetic toolbox for controlled gene expression.
