The promoter is a section of DNA sequence identified, bound, and initiated for transcription by RNA polymerase, located upstream of the transcription start site, which contains the conserved sequence required for RNA polymerase specific binding and transcription start, and the promoter itself is not transcribed.

This biobrick is an improvment of the biobrick BBa_I14018 designed by Group: Antiquity(2004)

Characterizing Part:

T--BIT--Figure Improve2.png

We modified the sequence of the promoter BBa_I14018, and got the promoter BBa_K2708009. Then we assembled these two promoter with reporter BBa_E0840 and transforming plasmid to Ecoli DH5α, measuring green fluorescence to characterize the effect of the promoter.

T--BIT--Figure Improve1.png

The figure below is the fluorescence results we measured in the eighth hour of culture. Excitation and emission are 480nm and 520nm, E0840 is the negative control. The results show that the promoter K2708009 has a stronger starting ability than I14018.
