A pTet promoter (can be repressed by tet Repressor, and transcription activity can be restored by addition of anhydrotetracycline (aTc)) produces transcripts of a spacer (including the RBS)-cro complex. The spacer is a temporary placeholder, where later cis sequences (for cis riboregulator) will be cloned in. This construct is an intermediate, as the terminator sequence for the transcript has not yet been cloned it.
