pPsiTac1
This part is a hybrid synthetic promoter composed of the -35 and the Pribnow box sequences of pTacI promoter (BBa_K864400) [1] and the consensus binding site of PsiR regulator of Rhizobiale [2].
Usage and Biology
D-Psicose inducible promoters naturally occur in nature, but they are quite long and not well characterized. In order to assess the efficiency of our psicose biosensor(s), we engineered a strong promoter, pTacI (BBa_K864400), regulated by LacI to make it psicose inducible.
pPsiTac1 is a synthetic promoter derived from pTacI (BBa_K864400) from which we removed the LacO sequence recognized by LacI and replaced it by the binding sequence of a transcription factor, PsiR (BBa_K2448006). PsiR is a LacI family transcription factor interacting with psicose, this is why the regulation of its promoter is similar.
As shown by the characterization of our psicose biosensor (BBa_K2448027), pPsiTac1 is strongly regulated by PsiR thanks to the consensus sequence, and the reporter gene shows high level expression under psicose induction. pPsiTac1 can therefore be categorized as a strong psicose inducible promoter.
