Sensor of halide+dehalogenase
RdhANP is a kind of dehalogenase first discovered in Nitratireductor.pacificus and it has been considered as one of the most promising enzyme to deal with organic halide in wastewater due to its unparalleled specialty working in oxygenic conditions. This year we plan to use RdhANP as the cleaner of the wastewater and try our best to make the management more suitable in reality. We get an inspiration from Peking-China’s work in 2013. In part BBa_J61051, they designed a circuit which can sense salicylate and induce the following reactions for its degradation. Through their results, we get the information that this sensor can also function when other substances like CL-PHL, which can serve as substrate of our enzyme RdhANP . Thus, we decide to make our brick which combines both RdhANP and BBa_J61051 and only when the substrates are detected, enzyme will be produced. It’s necessary to improve the accuracy of the project and release the burden of the bacteria. However, problem still remains. That is, this device has little capacity to resist the disturbance of other substances which can give the sensor a stronger massage to produce enzymes. Thus, only when the substrates of RdhANP occupy large proportion can this device exploit its advantage. And we plan to solve this problem by eliminating these interference before the management of organic halide using the device described by Peking-China to turn substance like salicylate into intermediates of TCA cycle.
