We designed this part to test our bacterial cell adhesion hypothesis. We predicted that SpyTag and SpyCatcher will form a covalent bond upon contact, which is what we desire in our gene circuit.

To test this visually, we express SpyCatcher on the surface of E. Coli with Intimin, a surface display protein. When the GFP-SpyTag fusion protein is added, we can observe the area around the cells to present green fluorescence as opposed to the control cells that were only exposed to the GFP protein without the SpyTag.
