FepA is an outer membrane receptor protein for Escherichia coli involved in the uptake of the ferric enterobactin siderophore. This part is a deletion mutant of the wild type FepA, which has been shown to convert the protein into an open diffusion channel, similar to a porin. It allows larger molecules through than normally would be permitted. The L8T mutant is otherwise similar to the L8 mutant apart from the introduction of a beta turn at the deletion junction (NCET).

The porins usually present will allow molecules up to around 700Da to freely diffuse in and out, however with this device, molecules of at least 1400Da should be able to pass through. Permeability was tested using the antibiotic Bacitracin. Wild type E. coli are resistant to this as the molecule is 1422Da in size. Expressing the device made the bacteria sensitive to bacitracin.
