This part is used for 2016 WM iGEM Ribozyme Characterization project. Lou et. al showed that sequences in the 5' UTR of a gene can affect the input-output relationship (transfer function) of a circuit. The RiboJ serves as an insulator to generalize the transfer function of a circuit regardless of promoter. The part is used in combination with BBa_K2066016 (constitutive Lac Repressor) to look at input-output relationship of IPTG-sfGFP. This part is the same as K2066014 but with a different promoter. The promoter for this part is J23116 as opposed to the plLacO used in K2066014.

This part is part of a library of parts designed to characterize the influence of RiboJ on the absolute gene expression level on various promoters, including the entire Anderson Promoter Library.

The part follows the UNS standard developed by William and Mary iGEM 2016, in which the insert is flanked by UNS 2 (BBa_K2066018) and UNS 3 (BBa_K2066019) for ease of cloning via Gibson or Golden Gate Assembly.
