Scaffold RNA that targets the region c6 of CYC1 promoter and recruits two times the protein repressor module PCP-Mxi via two effector protein recruitments site PP7. PP7 is a well-characterized viral RNA sequence which is recognized by PCP protein. This RNA is composed of the dCas9-binding sgRNA that interacts with CYC1, and two times the PCP-binding RNA hairpin, PP7.

Scaffold RNA with two PP7 hairpins that targets the region c6 of CYC1 promoter

Usage and Biology

A scaffold RNA is a guide RNA (gRNA), supplemented with one or more additional hairpins that can recruit different RNA binding modules. In this case, the hairpins are two times PP7, PP7 being a well-characterized viral RNA sequence which is recognized by PCP RNA binding module. This allows the recruitment of fusion proteins, like BBa_K1941000, to act on the level of expression of the promoter CYC1.
