Scaffold RNA that targets the region c3 of CYC1 promoter and recruits the protein repressor module MCP-VP64 via the effector protein recruitments site MS2. MS2 is a well-characterized viral RNA sequence which is recognized by MCP protein. This RNA is composed of the dCas9-binding sgRNA that interacts with CYC1, and the MCP-binding RNA hairpin, MS2.

A scaffold RNA is a guide RNA (gRNA), supplemented with one or more additional hairpins that can recruit different RNA binding modules. In this case, the hairpin is MS2, a well-characterized viral RNA sequence which is recognized by MCP RNA binding module. This allows the recruitment of MCP fusion proteins to act on the level of expression of the promoter CYC1.

scCyc1_MS2

Scaffold RNA with MS2 hairpin that targets the region c3 of CYC1 promoter.

Usage and Biology

A scaffold RNA is a guide RNA (gRNA), supplemented with one or more additional hairpins that can recruit different RNA binding modules. In this case, the hairpin is MS2, a well-characterized viral RNA sequence which is recognized by MCP RNA binding module. This allows the recruitment of MCP fusion proteins to act on the level of expression of the promoter CYC1.
