Detecting arsenic in Chinese medicine

Objective

     Measure the E. coli fluorescence intensity in different arsenic concentration with 3μg/ml Chinese medicine.

Methods

     Arsenic solution with different arsenic acid concentration (0, 1, 10, 50 and 100 ppm) is added into E. coli DH5α with Plasmids encoding GFP under arsenic promoter(part: BBa_K1106004).
     Fluorescence was measured at:

  • Excitation peak: 485nm
  • Emission peak: 538nm
  • Auto cutoff: 515nm

every 30 minutes.

Results

     This part is still specific to arsenic in Chinese medicine, so we can apply it in Chinese medicine examination.

Discussion

     Since E. coli expresses stronger fluorescence in higher arsenic concentration, this is a relevant promoter to detect the existence and measure the amount of arsenic.

Igem BsAs 2013

Objective Characterize the response of the arsenite sensitive promoter measuring GFP production upon different arsenite concrentrations.

Methods Several cultures carrying a plasmid that encodes GFP under this promoter(Bba_K1106004) were grown overnight with different arsenite concentration (0, 10, 50, 500 and 1000 ppb). After sonication, fluorescence was measured at 395nm excitation peak and 515nm emission peak. Each measure was normalized by culture density (OD 600nm).

Results As shown in the graphics below GFP production increases with the arsenite concentration.

Conclusion The promoter is sensitive to different arsenite concentrations.
