This part contains an additional BamHI restriction site at beginning of DAMP-Pex sequence and a XhoI restriction site between DAMP-Pex and double terminator(B0010-B0012). Also have a TEV Cleaveage site(ENLYFQS) between DAMP and Pex.
To achieve constitutive expression of this protein complex we put lacIq promoter at the beginning. It is a single base changed version of lacI promoter which boosts expression of gene about 10-fold. As this promoter cannot be terminated by T7 terminator like most of our parts we inserted double terminator (B0010-B0012) at the end.
