pompC and lacI box regulated cI expression
Uses a positively regulated promoter (pompC) and a lacI binding site to regulate the expression of cI. This construction was designed to be regulated by cph8 red light receptor (cph1-envZ chimeric protein). The purpose was to create a circadian switch to alternate the expression of two sets of genes.
Due to a lack of time we decided to have this construction synthesised instead of using the assembly protocol. Unfortunatly we did not succeed in cloning the final construct with the cyrcadian switch genes.
The integrity of our construct was checked by sequencing using VF2 primer and cI specific primers.
The aim of this construct was to create a circadian switch to alternate the expression of two sets of genes (Figure 2). This enables the production of one protein by night and another by day.
This regulation system is a NOT logic gate controled by a day/night.
