Using a fluctuation analysis mutation rate experiment, XL1Blue bacteria transformed with this construct yielded a mutation rate of 3.14E-09 mutations per cell per culture although mutation rate is dependent on growth conditions and can vary depending on experimental setup. This plasmid appears to incur a growth penalty in transformants.
