The DarR operator is a 14 bp palindromic DNA sequence discovered in ''Mycobacterium smegmatis''. The DarR operator can be bound by the HTH-transcription factor DarR. In ''M. smegmatis'', DarR and the DarR operator control the expression of the darR gene (auto-regulation) and other genes predicted to be involved in different processes. The sequence motif of the DarR operator varies slightly dependent on the downstream gene. (Zhang et al., 2013)

DarR operator

This part is the binding sequence of the c-di-AMP-dependent regulatory protein DarR. When bound by DarR, it is supposed to prevent the downstream information to be transcribed.
The important information: The operator is not in the pSB1C3 Vector but instead in the pBluescript-KS plasmid. This plasmid provides an ampicillin resistance. Furthermore, it has got a pUC origin.
