TapZ engineered membrane receptor

TapZ is an engineered membrane receptor capable of detecting several didpeptides.

Once activated TapZ phosphorylates the ompR protein which is then an active transcription factor.

Usage and Biology

In order to detect the S.Aureus we take inspiration on the parasite project ( http://2010.igem.org/Team:Imperial_College_London ) but there was one problem there is no peptide detector in E.coli who was able to activate a promoter. One receptor, involved in chemotaxis, detects few di/tri peptides (Tap receptor). In order to activate a promoter in presence of that peptide we designed a hybrid Tap/EnvZ receptor (which we called TapZ). EnvZ is a receptor involved in osmotic pressure sensing. It phosphorylates ompR which activates the pompC promoter. This part must be used in a ΔenvZ strain.
