We have characterized this part in three different chassis: TOP10 e.coli, Laboratory strain bacillus subtilis and a strain of bacillus subtilis with its normal fluoride riboswitch knocked out (kindly provided by the Breaker lab in Yale). Results of Miller assays for these three chassis are shown below.

This riboswitch is a positive regulator. The riboswitch, originally isolated from Bacillus cereus, serves as a transcriptional attenuator in the absence of fluoride. In the presence of fluoride its conformation changes and the repression is lifted. In B. cereus this serves to permit translation of a fluoride efflux pump, which allows the bacteria to cope with the potentially toxic fluoride levels in which it finds itself.

We have found, results in the experience tab, that the linear region of this riboswitch is in the 0uM to 100uM range when using &beta;-galactosidase as the reporter in a Miller assay, and in the 1mM to 30mM range when using &beta;-galactosidase as the reporter in an X-Gal assay.

Riboswitch that is highly sensitive to the F- ion. Usage and Biology

This riboswitch is a positive regulator. The riboswitch, originally isolated from Bacillus cereus, serves as a transcriptional attenuator in the absence of fluoride. In the presence of fluoride its conformation changes and the repression is lifted.
