Usage and Biology
This part is intended to bring a synthetic gene under the control of miR-34 in human cells. A gene carrying a target site for a functional miR will be repressed by RNA interference. The target site works by placing it downstream of the ORF you want to repress. In our hands, a gene with this part appears to be repressed in one cell line (U2OS) where miR-34 is produced at a minimal level in proliferating cells.
