Red light sensor with GFP reporter
Red light sensor with GFP reporter. For more information and references check the red light sensor motherpart (BBa_K568000). However, the data obtained did not match our expectations. We tested its function in JT2. The test of the assay was repeated several times. Our results indicate that the part is independed of the different light conditions active.
Therefore, testing or even the design of this part should be revised.
Experimental testing
The sensor was tested on pSB1C3 in BL21 DE3. After incubation over night under the on-light of 705 nm, off-light of 650 nm, in ambient light and in the dark. FLUO/ABS is a relative measurement of the GFP output as the fluorescence is divided through OD 600.
Another assay was set up to test the time dependency of the construct which did not alter our findings. In this experiment, the cells had been incubated over night in the dark and had been transferred into the different lighting conditions at t = 0 min. The construct yielded more GFP output than the constitutive promoter at all times.
