However, pT7 can only function under presence of associated regulatory protein; thus, for example, when we transform this construct into strain BL21 E. coli, which express associated regulator, both tetR and GFP will be expressed.In contrast, if we transform this construct into other strain, such as DH5alpha and S17-1,  both tetR and GFP genes will not be expressed. Due to that tetR and GFP are under the regulation of the same promoter, we can quantitize the tetR expression by detection intensity of GFP.
