In ''Synechocystis sp.'', this promoter transcribes CpcG2-phycobilisome as a chromatic acclimation system. Promoter pCpcG2 is regulated by the two component system CcaS/R. CcaS contains a histidine kinase domain whose autophosphorylation activity is up-regulated by green light. pCpcG2 promoter, inverted sequence (Green light inducible)

Inverted Green light inducible pCpcG2 promoter, from pJT122 plasmid constructed by Tabor et al. (2010) (1). It is positively regulated by the two component system CcaS/R, which exhibits a maximum response in 535 nm and is inactivated in 650 nm light (2). Light intensities must be carefully regulated to achieve successful gene expression. The sequence was inverted to ease DNA synthesis.

Usage and Biology

In Synechocystis sp., this promoter transcribes CpcG2-phycobilisome as a chromatic acclimation system. Promoter pCpcG2 is regulated by the two component system CcaS/R. CcaS contains a histidine kinase domain whose autophosphorylation activity is up-regulated by green light. When active, CcaS phosphorylates the cognate response regulator CcaR; phosphorylated CcaR then induces expression from pCpcG2 promoter (2).
