OL region from Lambda phage. Allows biphasic switch (K566002) effective repression: When placed around 2.4 kb apart from OR (included in pRM) and in high [cI] conditions, cI octamerizes and loops the DNA stabilizing cI binding to OR3; such structure represses pRM promoter (K566001).

Usage and Biology

OL is a set of three binding sites: OL1, OL2 and OL3. It makes part of a well studied regulatory region in Lambda (1). Its binding sites maybe used to design new regulatory networks or to mimic Lambda's natural behavior. The biphasic switch (BBa_K566002) makes a good example of that.
