Talk:Part:pSB1AC3
We could not confirm that bacteria were resistant up to 3500 ug/ml chloramphenicol as initially reported. Our experiments were performed with DH5alpha harboring pSB1AC3 in LB medium containing different concentrations of chloramphenicol. In the first scan, we identified the limiting concentration to be around 1 mg/ml. Next, we tested as many as 19 different concentrations (between 50 and 4000 ug/ml) according to a recently published protocol (Wiegand et al, Nat Protoc, 2008) and determined the minimal inhibitory concetration to be 800 ug/ml. This work was done by Nina Erčulj and Nives Škrlj in [http://openwetware.org/wiki/FCCT_Biochemistry_Lab our lab] in Ljubljana in March 2008. (MDolinar)
