Design

The actuator receives a PoPs signal in order to produce melamine deaminase via the modified TriA gene. Its design comprises three components, which include a bicistronic ribosome binding site (apFAB682), the MDA gene of interest which codes from melamine deaminase, and a transcription terminator (apFAB376). The ribosome binding site was chosen to have a moderately high performance and a relatively small deviation so as to not overburden the cell with production of melamine deaminase and to keep deviation to a minimum while still promoting heavy production. The terminator was chosen to have a high percentage of termination to make sure that ribosome interaction was being finalized at the end of our actuator.
