1. The DNA level The plasmid PET-Phok was selected for verification, and the transformed plasmid DH5a (phoK) was verified by plasmid running gel and PCR. As can be seen from Figure 1, the sample band was over 5000bp, basically in line with the target band of 7863bp, which proved that our plasmid transformation was successful.
We also carried out enzyme digestion verification on the plasmid. Marker was 10000bp. Sample 1 was the target plasmid, and sample 2 was the sample with endonuclease Smal and EcoRl added for enzyme digestion. There was also a smaller band between 5000bp and 6000bp, which was consistent with the target band 5239bp obtained by enzyme digestion, which further indicated that our plasmid transformation was successful.(Figure 2)
