miR-21 sensor
The expression of miR-21 was found to be significantly associated with all tumor stage of gastric cancer. We designed miR-21 sensor containing two complementary binding sites to miR-21, which inhibit miR-21 expression. The sequence of miR-21 sensor was synthetised by GenScript (Shanghai, China). We inserted miR-21 sensor into the 3' end of Cherry, which generated Cherry-miR-21 sensor. The GFP-miR-21 sensor was controlled by CMV promoter (CMV-Cherry-miR-21). The plasmids of CMV-Cherry-miR-21 were transfected into different cell lines (one is human gastric epithelial cell line. The others are gastric cancer cell lines).We also measuresd the fluorescence of GFP in different cells transfected with CMV-Cherry-miR-21 for 24 h by plate reader (SpectraMax i3), which analyses the potential value of miR-21 sensor to measure the expression of miR-21.
