This promoter comes from rhizobial phage 16-3, and was engineered to be inducible by anhydrotetracycline (aTc). The sequence was taken from Chubiz et al. and shortened it to 60 base pairs and then added the GGA appropriate sticky ends. The E coli cells were placed in LB+amp media with differing cell conditions, including our experimental promoter, promoter part:BBa_J04450 as a positive control, and buffer with no promoter as a negative control. All these conditions were either exposed to aTc or not exposed, on separate plates. This part works as a promoter, and performance was enhanced in the presence of aTc.
