The original BioBrick standard does not support in-frame assembly of protein domains. However, several new standards have been developed that do support in-frame assembly of protein domains. In particular, Assembly standard 23 (the Silver standard) and Assembly standard 25 (the Freiburg standard) both support assembly of protein domains and are backwards-compatible with the original BioBrick assembly standard.
