Robotic assembly uses these files to control the robot based on the desired assemblies.


Stage-n-picking.csv

  • The user specifies the assemblies to perform. The program selects plasmid DNA for the left and right inserts.
  • picking.csv moves 5 ul of the part's plasmid DNA from the library plates to the correct wells on the digest plate.


Stage-n-c-plasmids.csv

  • The program, determines how much of each construction plasmid will be required and lays out the construction plasmid digest wells.
  • c-plasmid.csv moves 5 ul of construction plasmid from the reagent plate to the construction plasmid digest wells.

Stage-n-mixing.csv

  • The robot will move digest well contents from the right side of the digest plate to the corresponding well on the left side of the digest plate.
  • mixing.csv moves 10 ul of cut construction plasmid to the correct digest well on the left side of the plate for mixing and ligation.


Stage-n-dilution.csv

  • Each serial dilution happens in broth with the correct antibiotic. This is staged in the Antibiotic Broth plate.
  • dilution.csv fills the Antibiotic Broth plate with broth containing the correct antibiotic from the broth resevoirs.
  • The single colony results will later be grown in the Antibiotic Broth plate filled with broth using this file again.
