Kanamycin Resistance for Synechococcus
Confers resistance to the antibiotic kanamycin. This sequence was used within the plasmid BBa_K4047039. This plasmid, which deletes the target gene glpX by insertion of kanamycin, simultaneously using this resistance-coding gene (KanR) as a selective marker for successful transformation.
Colonies were grown on BG11 media containing kanamycin. PCR was used to verify the change in genomic content, with an increasing band width indicating successful integration of the kanamycin resistance gene into the target site. This part is an improvement to the documented part BBa_K1424003, which was optimized for Synechocystis sp. PCC 6803. This has been optimized here for use in the organism Synechococcus elongatus PCC 7942, and as such has a different DNA sequence.
