mEmerald
mEmerald is a derivative of the Green Fluorescent Protein from Aequorea victoria with improved features in terms of intrinsic brightness, photostability, resistance to bleaching, maturation and folding at 37°C [1]. The protein contains several mutations compared to wild-type (Uniprot P42212): +2V, F64L, S65T, S72A, N149K, M153T, I167T, A206K and H232L.
Figure 1 illustrates the intense colour of mEmerald expressing E. coli cells.
Usage and Biology
Applications are various: Fusions, promoter & secretion studies, FRET and other quantitative imaging techniques.
Our team used this codon optimized mEmerald to build the Universal Biosensing Chassis (BBa_K2448023).
