BBa_K1909007 is a construct of strong Anderson promoter BBa_K081005, a strong ribosome binding site and the receptor mTaz, part BBa_K1909002, capable of detecting meso-2,6-Diaminopimelic acid (mDAP). The receptor mTaz is based on the chemotaxis receptor Taz, part BBa_C0082, capable of sensing aspartate and in combination with the cytosolic domain of osmosensor kinase EnvZ.
Usage and Biology
This combination has the highest expression of the three combinations BBa_K1909007, BBa_K1909008 and BBa_K1909009.Testing the combination showed that cells grew very slowly (for miniprep culture more than 48 hours). We assume that this combination (strong promoter and strong RBS) induces production of toxic amounts of the transmembrane receptor.
