rtTA is a regulatory protein which can combined with tetracycline analogues.

rtTA is a regulatory protein which can combined with tetracycline analogues. Like the protein in Biological signaling pathways. When it combine with tetracycline analogues, it can change its conformation to activate the subsequent signal path. Under the action of constitutive promoter, the regulatory protein expresses stably. When it combine with tetracycline analogues, rtTA change its conformation to activate the promoter, then the report gene expresses.

In the pTet-on, the part plays a role in regulating the protein. This part is the switch of tetracycline in the Recombinant yeast cells. It can ensure the system can work well and are related to the sensitivity of the detector. Under the action of constitutive promoter, the regulatory protein expresses stably, then play the role of switch.We get the pTet-on plasmid from biowit company and the design primers to get the segment by PCR.
